Organic molecules called photoswitches (PS) can become fluorescent on the exposure to light or chemical reaction and revert to the initial state, which is non-fluorescent, on illumination with a different light or chemical. Thus, they have found wide applications in chemosensors. However, in many cases, the applications suffer due to the slow response time of the sensors towards the target molecules. An ultrasensitive fluorescent chemosensor with a fast response can be of a great help in molecular recognition. We have developed a three-component ruthenium(II)-based photoswitching probe, on the basis of picolinamide derivative, for the ultrasensitive recognition of biomolecules such as DNA, proteins and other metabolites. Our goal was to design a solution-based photoswitching approach that is suitable for quantitative recognition of various biomolecules. Thus, we have developed (a) highly sensitive turn-on fluorescent sensor (b) for the identification of target biomolecules through fluorescence quenching, on the basis of our photoswitching probes and (c) for the identification of biomolecules using multi-channel monitoring. Our studies indicate that the designed probe can detect various biomolecules in the nano-molar concentration, through fluorescence quenching and fluorescence turn-on sensing. Thus, the present study may help in understanding the mechanisms of the photoswitching probes towards the target biomolecules and make them suitable for applications in molecular recognition as well as in the protein and nucleic acid detection in medical and biological fields.
